Melanotan 2 vs PT-141: Sexual Function and Tanning Peptides Compared

Melanotan 2 (MT-2) and PT-141 (Bremelanotide) are both melanocortin receptor agonists. MT-2 produces tanning, sexual arousal, and appetite suppression. PT-141 is a metabolite of MT-2 that is FDA-approved for HSDD in women and focuses on sexual arousal without tanning. PT-141 has a better safety profile for sexual health applications.

The Melanocortin System

Both Melanotan 2 and PT-141 work by activating melanocortin receptors (MC1R through MC5R). These receptors regulate diverse functions including skin pigmentation (MC1R), sexual arousal and erection (MC3R, MC4R), appetite and metabolism (MC4R), and inflammation (MC1R, MC3R). The melanocortin system is a key regulator of both sexual function and body composition.

Melanotan 2: The Tanning Peptide

Melanotan 2 (MT-2) is a synthetic cyclic analog of alpha-MSH that activates all melanocortin receptors. Its effects include: skin tanning (via MC1R stimulation of melanocytes), sexual arousal and spontaneous erections in men (via MC3R/MC4R), appetite suppression and fat loss (via MC4R), and nausea (a common side effect). MT-2 is not FDA-approved and carries risks including activation of existing moles and nevi, which has raised concerns about melanoma risk with long-term use.

PT-141 (Bremelanotide): The Focused Sexual Health Peptide

PT-141 is a metabolite of MT-2 that was developed specifically for sexual dysfunction. It activates MC3R and MC4R (the receptors responsible for sexual arousal) without the strong MC1R activation that causes tanning. PT-141 is FDA-approved as Vyleesi for hypoactive sexual desire disorder (HSDD) in premenopausal women. For men, it is used off-label for erectile dysfunction and low sexual desire. Typical dosing: 1–2 mg subcutaneously 1–2 hours before sexual activity.

Key Differences

Tanning: MT-2 causes significant tanning; PT-141 causes minimal tanning. Sexual effects: both produce sexual arousal; PT-141 is more selective. Nausea: both can cause nausea; MT-2 more so. Safety: PT-141 has a better characterized safety profile and FDA approval. Regulatory status: PT-141 is FDA-approved (Vyleesi); MT-2 is not approved anywhere. For sexual health applications, PT-141 is the preferred and safer option. MT-2 should be used with caution given the melanoma risk concerns with long-term use.
